I only got one key with my 03 Prius so I took it to the dealer the next week to get a replacement key. They took the key, the car and half and hour later, charged me $65!

The maintenance manual and several web pages have the instructions on how to program the engine controller to recognize another key. You can buy them on Ebay for about $5 and having a backup is a good thing.

Our 01 has anti-theft and horn-honking style alarm, the key has a chip that needs to be present for the car to start. Very difficult to make one run unless you have the key.
